Non-secreted viruses are viruses which reproduce within the cultured cells. Since the viruses are contained within host cells, the cells themselves must be harvested and processed in order to harvest the viruses that lies inside.
If the cells are mixed with the culture medium (which is the case for stirred tanks), the culture broth would require extensive downstream processing. First requirement would be to separate the virus-containing-cells from the culture medium and the other components are mixed in it.
After the cells are harvested, methods such as freeze and thaw can be implemented in order to open up the cell and harvest the virus inside. After harvesting the virus, further downstream processing would be required in order to purify, clarify and concentrate the virus.
